Protein A Affinity Resins

  • Protein A affinity resin is the most cost-intensive critical material in the downstream purification of antibody drugs, primarily used for capturing target antibodies from fermentation broth. Protein A, derived from Staphylococcus aureus, binds specifically to proteins containing the Fc region. By optimizing and engineering the Protein A structure to enhance alkali resistance, it is widely applied as a functional ligand in affinity chromatography resins. 

    Sepax Technologies has developed agarose-based MabPurix A series affinity resins and PMA-based MabPurix P45 affinity resin. Both series have achieved large-scale production with stable supply and consistent quality.

MabPurix A65 Excel 

  • The MabPurix A series affinity resins are based on spherical, narrow-dispersity, highly crosslinked agarose gel matrices. Compared with conventional agarose gels, MabPurix A resins offer improved pressure tolerance, allowing for higher column bed heights and greater batch processing capacity. Through Sepax’s proprietary technology, the agarose matrix is coupled with an alkali-tolerant recombinant Protein A ligand, specifically designed for the purification of mAb, BsAb, polyclonal antibodies, and Fc-fusion proteins.


MabPurix P45 Affinity Resins 

  • The MabPurix P45 affinity resin is based on a polymer microsphere matrix with an average particle size of 45 μm. It is suitable for applications requiring high mechanical strength and fast flow rates. Through Sepax Technologies’ proprietary technology, the polymer matrix is coupled with alkali-resistant recombinant Protein A ligand, specifically designed for the separation and purification of antibody drugs such as monoclonal antibodies, bispecific antibodies, multispecific antibodies, and Fc fusion proteins, especially high binding capacity for high-molecular-weight antibodies. The media surface has been treated with proprietary technology to achieve better hydrophilicity, maximally avoiding non-specific adsorption with biological samples.

mRNA Affinity Resins  

  • Most RNA molecules contain a polyadenylate tail (Poly A tail) with a length of up to 250 bases. According to base pairing principles, T base (thymine) molecules can form base pairs with A bases. Based on this principle, oligo(dT)20 affinity chromatography media has been developed for mRNA purification, with the ligand being oligo dT20, used for high-efficiency separation and purification of mRNA samples containing Poly A tails. 

    The dT20 affinity media developed by Sepax Technologies includes Proteomix POR50-dT20 based on polystyrene/ divinylbenzene matrix, as well as Monomix dT20 based on polymethacrylate matrix, which can be used to capture and purify mRNA from IVT transcription reaction solutions.
